Abbreviations MLC micellar liquid chromatography FDA food and drug analysis 2 Abstract A simple and robust method was developed for the routine identification and quantification of amoxicillin by micellar liquid chromatography. Amoxicillin, a β-lactamase inhibitor, is one of the most commonly prescribed drugs in the treatment of urine and skin structure infections. In this work, amoxicillin was determined in urine samples without any pre-treatment step in a phenyl column using a micellar mobile phase of 0.10 M sodium dodecyl sulfate and 4% butanol at pH 3. A UV detection set at 210 nm was used. Amoxicillin is eluted at 5.1 min with no interference by the protein band or endogenous compounds. Linearities (r > 0.9998), intra-and inter-day precisions were determined (RSD (%) 0.4-2.7% and 0.3-5%, respectively in micellar media, and 0.14-2.6% and 0.13-6%, respectively in urine), and robustness was studied in the validation of the method. LOD and LOQ were 0.06 and 0.4 µg/mL in micellar media and 0.11 and 0.4 µg/mL in urine, respectively. Recoveries in the urine matrix were in the range of 95-110%. The validated method proved to be reliable and sensitive for the determination of amoxicillin in urine samples.

Optimized Method for Determination of Amoxicillin, Ampicillin, Sulfamethoxazole, and Sulfacetamide in Animal Feed by Micellar Electrokinetic Capillary Chromatography and Comparison with High-Performance Liquid Chromatography

Antibiotics as additives in animal feedstuffs are forbidden in many countries in the world, but they are still abused. A micellar electrokinetic capillary chromatography method was performed at 25 C and 30 kV (under pressure 15 mbar) using 25 mmol dm phosphate buffer (pH = 8.0) containing 70 mmol dm sodium dodecysulfate (SDS) and 10 % (volume fraction) methanol as the background electrolyte fo...

Separation of Sedative – Hypnotic Drugs with Mixed Micellar Liquid Chromatography

Separation of ten sedative- hypnotic drugs was performed by RP-HPLC using mixed micellar mobile phase. Effect of temperature, type and amount of organic modifier in mobile phase on efficiency (N) and asymmetry factor (B/A) showed that, the appropriate conditions for a good separation were 35°C and 7% (V/V) butanol in mobile phase.  Variations of selectivity factor versus butanol concent...

Use of Temperature-controlled Ionic Liquid-assisted Dispersive Liquid-Liquid Microextraction Method for the Detection of Amoxicillin, Cloxacillin and Erythromycin Residues in Cow Milk using High Performance Liquid Chromatography

Background and Objectives: Different types of antibiotics are used indiscriminately in animal husbandry for the growth and treatment of livestock, which cause accumulation of antibiotic residues in livestock products such as milk. Long-term consumption of milk contaminated with antibiotic residues above the permissible limits is harmful to human health, causing antibiotic resistance problems. T...

Development and Application of a Validated Liquid Chromatography-Mass Spectrometry Method for the Determination of Dexchlorpheniramine Maleate in Human Plasma

A convenient liquid chromatographic-single Quadrupole mass spectrometric (LC-MS) method was developed and validated for dexchlorpheniramine maleate (INN name: chlorphenamine) determination in human plasma. The need for just a single liquid-liquid extraction with ethyl acetate and being highly sensitive were the advantages of this method. The linearity was also excellent over the range of 1...
